- During the development and homeostasis of multicellular organisms the balance between proliferation and quiescence needs to be precisely coordinated. Key to this balance are dynamic changes in the expression of cell cycle proteins. To visualize the status of the cell cycle machinery during the transition into quiescence in living cells we employ homologous recombination to insert genes of fluorescent proteins site-specific the loci of crucial cell cycle regulators. Using quantitative live imaging and automated image analysis approaches we extract total and relative levels of key cell cycle regulators from single cells to obtain a more quantitative view the decision between proliferation and quiescence.
