 in-hindbrain boundary (MHB) acts as a lineage restriction boundary and is 
 thought to prevent the intermingling of prospective midbrain and hindbrain
  cells during embryonic development. The MHB also provides\, as an organiz
 er cell population\, positional information to the neighbouring cells\, wh
 ich take different lineage decisions. The aim of the proposed project is t
 o identify the molecular regulators that mediate cell sorting in the devel
 oping MHB. To achieve this\, first\, the role of differential cell-cell ad
 hesion or surface tension in cell sorting at the MHB is studied using atom
 ic force microscopy and live imaging techniques. Second\, the molecular me
 chanisms controlling cell sorting and tissue border formation is addressed
  using zebrafish mutants of a candidate-signalling pathway. Finally\, the 
 role of cell sorting in lineage specification is addressed using lineage-t
 racing analysis in combination with inactivation of genes affecting cell s
 orting at MHB. The results obtained from this study will increase our unde
 rstanding of the fundamental process of boundary formation and patterning 
 of the embryo.
