 ption: Enhancers can regulate their target promoters over large genomic di
 stances for which spatial proximity between these elements is assumed to b
 e necessary. Topologically Associating Domains (TADs) are thought to provi
 de a spatial environment in the nucleus\, harboring multiple enhancers and
  are thus related to gene regulation. However\, to which degree TADs contr
 ibute to gene regulation and how enhancer position within the TAD affects 
 its function in vivo is not known.  I will present work that uses genome-e
 ngineering to generate a series of mouse lines with alleles designed to te
 st how important TADs are for gene regulation. At well-characterized devel
 opmental loci\, we either re-arrange TAD boundaries or reposition individu
 al enhancers and analyze the effects on 3D chromatin structure\, gene expr
 ession and mouse phenotype in vivo. Thereby we can show that TADs provide 
 a supportive environment for enhancer function but also point to our limit
 ed understanding of the interplay between regulatory elements.
