The analysis of biomolecular interactions, such as protein-protein, protein-nucleic acid or protein-small molecule, is a prerequisite to understand biological processes on a molecular level. In the recent years MicroScale Thermophoresis (MST) has proven to be a fast and efficient tool for the quantification of biomolecular interactions. As MST yields important information about sample quality such as aggregation in addition to the dissociation constant, this method allows for a straight forward optimization of sample conditions. In order to get the maximum out of an MST experiment, it is necessary read the signs of different events and artifacts. This presentation focuses on the practical aspects of MST such as experimental design, data evaluation and assay optimization. In detail subjects like non-standard binding events, alternative labeling strategies and fluorescence effects will be discussed.
