- Our body consists of various histological structures. These structures are generated via complex cell-cell interactions, but actual mechanisms of these pattern formation remain to be elucidated. Some cell culture systems retain the ability to generate histological structure, and they have been very useful in understanding various pattern formation phenomena in vivo. We utilize mathematical modeling to understand these phenomena, and in this talk we will present two examples. • MDCK Wound healing Compactly packed cells tend to retain their optimal distance, but the detail of the interaction remain to be elucidated. In wound healing assay of MDCK cells, we observe transmission of compression wave during the repair process. We could reproduce this phenomenon only by spring-mass system, in which cell-cell interaction influences the velocity of the cell. However, physically inertia should be negligible in this spatial scale, so the modeling predicts that some biological mechanism exists which is equivalent to inertia. • HUVEC pattern formation Human Umbilical Vein Endothelial Cells (HUVECs) has been known to retain an ability to form meshwork when cultured on appropriate extracellular matrix. Several attempts has been made to model this phenomenon, and these models falls into two classes. One utilizes mechanical property of extracellular matrix, and the other include vascular endothelial growth factor (VEGF) as a chemoattractant. We are now measuring VEGF diffusion dynamics in this culture system to determine whether chemotaxis toward VEGF gradient is the mechanism to generate meshwork pattern.
