- Transient interactions govern the interplay of biomolecules in virtually all processes of life. The analysis of molecular interactions is key for the discovery of new drugs and binding parameters such as affinity, avidity, and kinetic rates are helpful to select the most promising candidates during drug development. I will describe our attempts to develop new methods and devices to measure biomolecular binding with surface biosensors, single-cell biosensors, and nanopores for affinity-selective single molecule detection. I will highlight applications concerning new drug modalities involving multi-specific small molecules and antibodies, and real-time binding measurements on living cancer cells.
