Predict cell transition dynamics from the landscape constructed from single-cell data

- Although a gene is considered as the unit of structure and function of life, biological functions usually arise from the emerging properties of a complex gene regulatory network, which manifests as a “Waddington landscape”. However, it is difficult to elucidate all relevant gene interactions, build mathematical models, compute the landscape and cell dynamics consequently. With the rapid development of single-cell technology, we can easily measure the transcriptomes of thousands of cells simultaneously, which make it possible to compute the landscape directly from single-cell data. As proof of principle, we studied the landscapes to explain how melanoma cancer cells transition to resistant states from single-cell data using this method. The ultimate goal is to apply this method to find new cancer treatment against the drug resistance.
